Overview

When viewing a variant, the page displays different sections depending on the clinical significance tier. This ensures that clinically relevant information is prominently displayed for actionable variants, while less relevant details are hidden for benign or unknown significance variants.

Tier Definitions

Tier I - Actionable

Tier I-A: Biomarkers that predict response or resistance to FDA-approved therapies for a specific type of tumor, or biomarkers included in professional guidelines.

Tier I-B: Biomarkers with strong clinical significance based on well-powered studies with consensus from experts in the field.

For Tier I variants, the page displays:

  • Full clinical evidence including FDA-approved therapies
  • NCCN practice guidelines
  • CKB Genomenon therapeutic evidence
  • CIViC evidence
  • Relevant clinical trials

Tier II - Potential Significance

Biomarkers with potential clinical significance based on emerging evidence, CKB Level 5 evidence, or guidelines applicable to other tumor types.

For Tier II variants, the page displays:

  • NCCN guidelines for other tumor types
  • CKB therapeutic evidence

Tier III - Unknown Significance

Variants of unknown clinical significance with insufficient evidence for therapeutic actionability.

Tier IV - Benign

Variants classified as benign or likely benign with no clinical action recommended.

For Tier IV variants, the page displays:

View Name Display

The page header displays the somatic view name corresponding to the detected tier:

  • Tier I - Actionable: For Tier I-A and I-B variants
  • Tier II - Potential Significance: For Tier II variants
  • Tier III - Unknown Significance: For Tier III variants
  • Tier IV - Benign: For Tier IV variants

This badge helps users quickly identify the clinical significance category of the variant they are reviewing.

Manual Classification

All tiers allow manual AMP/ASCO/CAP classification. This enables clinical experts to:
